I have a customer who is at the other end of the country looking to have a design at 2500mm x 1300mm digitally printed.

Not usually a problem however the file size is proving too large to be e-mailed. (circa 50MB!) Full of high res photographs, which is the problem.

Designed in illustrator, tried sending as PDF, EPS and the .AI file however all are too large.

How can they optimize it to suit being e-mailed?

Get them to Jepg it on a compression setting of 10. This will be fine to print from and it will only be a couple of meg. This is how I get clients to send very large files to me if they cant use my ftp site or yousendit / mybigfile.
